ABSTRACT

Objective To observe the effects of nonylphenol(NP)on the intracellular calcium concentration changes and cell proliferation,and the involvement of GPR30 receptor in H9c2 cell. Methods The intracellular calcium concentration changes were recorded by using intracellular calcium determination method and cell proliferation was observed by MTT method in H9c2 cell. Results NP(1×10-10 mol/L)increased the intra?cellular calcium concentration changing amplitude and promoted the proliferation of H9c2 cells,while NP(1×10-6 mol/L)decreased intracellular calcium concentration changing amplitude and suppressed cell proliferation. G15 could block the promoting effect of 1×10-10 mol/L NP on the intracel?lular calcium concentration and cell proliferation,but could not block the inhibition of 1×10-6 mol/L NP on the intracellular calcium increase and cell proliferation. Conclusion The results indicate that NP affect rapid calcium signal changes and cell proliferation in non?monotonic dose dependent manner,and its mechanism may be due to the different involvement of GPR30 receptor in different concentrations.

ABSTRACT

Objective To discuss the clinical value of united detection of myocardial enzymes and troponin in the diagnosis of hypothyroidism.Methods 27 patients with hypothyroidism were chosen as the observation group.26 healthy volunteers were selected as the control group.CK-MB,LDH,AST,FT3,FT4,TSH,cTnT,cTnI were detected.Results The levels of serum AST,LDH,CK-MB of the observation group were significantly increased,the mean value was(52.67 ± 7.32)U/L,(265.23 ± 101.34)U/L,(26.75 ± 18.75)U/L respectively,which were significantly higher than the control group,the differences were statistically significant(P < 0.05).The c TnT and c TnI positive rate of patients in observation group were 70.4% and 81.5%,compared with the control group,the rates were much higher (P < 0.05).Conclusion The united detection of myocardial enzymes and troponin had high clinical value in the diagnosis of hypothyroidism.

ABSTRACT

Objective To observe the effects of nonylphenol(NP) on L-type Ca~(2+) currents(I_(Ca-L)) in isolated guinea-pig ventricular myocytes.Methods The ventricular myocytes were isolated from guinea-pig hearts.The L-type Ca~(2+) currents in the ventricular myocytes treated with NP were measured with whole cell patch-clamp technique.Results Nonylphenol inhibited lew,at different potentials.Nonylphenol (10~(-6) mol·L~(-1),10~(-5) mol·L~(-1)) reduced the peak amplitude of I_(Ca-L) from-3.2±1.5 pA·pF~(-1) to-1.6±0.8 pA·pF~(-1)(P<0.01) and-1.4±0.7 pA·pF~(-1)(P<0.01),respectively.Nonylphenol did not influence the activation curve of I_(Ca-L) significantly.Conclusion Nonylphenol could in hibit the L-type calcium currents in isolated guinea-pig ventricular myocytes with a concentration-dependent manner.Objective To observe the effects of nonylphenol(NP) on L-type Ca~(2+) currents(I_(Ca-L))in isolated guinea-pig ventricular myocytes.Methods The ventricular myocytes were isolated from guinea-pig hearts.The L-type Ca~(2+) currents in the ventricular myocytes treated with NP were measured with whole cell patch-clamp technique.Results Nonylphenol inhibited lew,at different potentials.Nonylphenol hibit the L-type calcium currents in isolated guinea-pig ventricular myocytes with a concentration-dependent manner.

ABSTRACT

Objective To investigate the molecular mechanism of CCl_(4) inducing fibrosis in hepatic stellate cells.Methods The expression of Transforming growth factor-?1(TGF-?1) at mRNA and protein level were detected by RT-PCR method and Western blot analysis.The proliferation and cell cycle distribution were detected with MTT method and flow cytometry.Results After treatment by(10 mmol/L) CCL_(4) for 24 hours,the mRNA and protein expression of TGF-?1 in CCl_(4) group sharply increased.In the meantime,the proliferation of CCl_(4) group cells was markedly increased.There were less G_(0)/G_(1) phase_() cells and more G_(2)/M phase cells in CCl_(4) group than those in control group.Conclusion Suitable CCl_(4) may increase the expression of TGF-?1 and the proliferation of hepatic stellate cells,which is one of mechanisms leading to hepatic fibrosis.

ABSTRACT

Aim To observe the effect of kanamycin on the change of Ca2+ concentration in outer hair cells caused by acetylcholine to explore the relationship between kanamycin ototoxicity and the cochlear efferent nervous system.Methods After the guinea pigs were killed by decapitation,the OHCs were isolated in artificial perilymph and divided into three groups:control group,acetylcholine(ACh) group and kanamycin + ACh group.OHCs were loaded with Fluo-3/AM for 30 min and then the change of Ca2+ fluorescence intensity was observed.Results Ca2+ fluorescent intensity in OHCs remained basically unchanged in control group,increased 3.06 times(n=6) in ACh Group,and increased 1.75 times(n=6) in kanamycin +ACh Group.Conclusions Kanamycin can obviously inhibit the increasing of Ca2+ concentration in OHCs caused by ACh.
